Protective Effects of Wolguk-whan Methanol Extract on Oxidative Injury in Cultured Rat Liver Cells


Abstract
Objectives : Wolguk-whan has been used for the treatment of stress, digestive system disease. This study was purposed to investigate the effects of Wolguk-whan methanol extract(WGWM1 on oxidative damage in cultured normal rat liver cell.
Methods : The levels of lipid peroxide(Lp0) were determined by TBA method in the linoleic acid autoxidation system. The scavenging effect of WGWM on a,a-diphenyl-P-picrylhydnuyl radical(DPP H ¡¤) was determined according to the method of Hatano. In the Fenton system(ferrous ion reaction with hydrogen peroxide), the levels of hydroxyl radical induced LPO in rat liver homogenate were determined according to the method of TBA. Inhibitory effect of WGWM on superoxide generation was measured by xanthine-xanthine oxidase system. Cultured normal rat liver cell(Ac2F) were prepared and incubated with or without WGWM. After 18hr, cells placed in DMEM medium without serum, and then incubated with 1mM tert-butyl hydroperoxide(tert-BHP) for 2hr. Viable cells were detected by MTT assay.
Results : In the linoleic acid autoxidation system, WGWM significantly inhibited the time course of the lipid peroxidation. WGWM showed about 86% scavenging effect on DPPH radical. And WGWM inhibited the lipid peroxide formation in rat liver homogenate induced by hydroxyl radical derived from Fenton system. And WGWM exhibited dose-dependent inhibitory effect on superoxide generation from xanthine-xanthine oxidase system. Cell killing was significantly enhanced by addition of tert-BHP compared to those of untreated group. WGWM pretreated cell resisted the toxic effects of tert-BHP.
Conclusions : These results suggested that WGWM might protect against lipid peroxidation by free radicals, oxidative injury of hepatic cell.
